【摘要】:ThisEuropeanStandardspecifiesamethodforthedeterminationofthemassfractionofniacininfoodstuffsbyhighperformanceliquidchromatography(HPLC)bythreedifferentwaysofhydrolysis,acidhydrolysis(A),enzymatichydrolysis(B)oracid/alkalinehydrolysis(C).Themethodhasbeenvalidatedininterlaboratorytestsonfortifiedandnon-fortifiedsamplessuchasbreakfastcerealpowder,chocolatecereals,cookedham,greenpeas,lyophilizedgreenpeaswithham,lyophilizedsoup,nutritiveorangejuice,milkpowderandwheatflour,atlevelsfrom0,5mg/100gto24mg/100g.Forfurtherinformationonthevalidationdata,seeAnnexB.AandBgivesimilarresultsforniacin.InoptionsAandBniaciniscalculatedasthesumofnicotinamideandnicotinicacid,andexpressedasnicotinicacid[1].OptionCgiveshigherresultsthanAandBforniacinwithnon-supplementedcereals,butsimilarresultsforotherproducts.InoptionC,niaciniscalculatedandexpressedasnicotinicacidaftertransformationofnicotinamideintonicotinicacid[2].OptionAisfasterandcheaperthanBandC.OptionBisusedifanexactquantificationofnicotinamideandnicotinicacidisneeded.ThiscannotbedonewithoptionA,becausethereisaslighttransformationofnicotinamideintonicotinicacidduringtheacidhydrolysis.OptionCquantifiestotalniacin.Thealkalinehydrolysisisabletoliberateotherformsgivinghigherresultsforniacin,whichinsomefoodssuchasmaizeandcerealsarenotnormallybiologicallyavailable,see[3],[4]and[5].InformationonacomparisonbetweenthethreedifferentwaysofhydrolysisisgiveninAnnexC.
